Furthermore, we also found CS activity that seemed to predict the upcoming events. We demonstrate that, in addition to carrying error-related information, CSs carry information on the metrics of both primary and small corrective saccades in a time-specific manner, with changes in CS firing probability coupled with changes in CS duration. When faced with the bewildering diversity of findings and views unraveled by highly specific tasks, one may wonder if there is just one true function with all the other attributions wrong? Or is the diversity of findings a reflection of distinct pools of PCs, each processing specific streams of information conveyed by climbing fibers? With these questions in mind, we recorded CSs from the monkey oculomotor vermis deploying a repetitive saccade task that entailed sizable motor errors as well as small amplitude saccades, correcting them. While initially thought to be specialized in reporting information on motor error for the subsequent amendment of behavior, CSs seem to contribute to other aspects of motor behavior as well. While there is consensus that SSs convey information needed to optimize movement kinematics, the function of CSs, determined by the PC’s climbing fiber input, remains controversial. ![]() ![]() There’s a bunch of exposition to explain the increasingly murky concept of time in the Rudolph-verse, but basically Happy has to be found before New Year’s Day or else the current year will go on forever. Easily the weirdest of the Rankin/Bass specials, Rudolph’s Shiny New Year is a mess: Taking place immediately after the first Rudolph, it features a time-traveling quest in which our favorite red-nosed reindeer travels across time periods (technically, time islands) to find Happy, the big-eared New Year’s baby that’s gone missing. If you watch the original Rudolph and think to yourself, “This is fun and all, but I really wish they found a way to work in a caveman, a camel-clock hybrid and Benjamin Franklin,” then this is the Christmas special for you. Thankfully, Paste is here to shepherd you through the world of Rankin/Bass, from worst to best. It can be difficult to know where to get one’s Yuletide on with such a massive catalogue. While there are Rankin/Bass entries for Easter and Thanksgiving, too, the company earned its reputation with a series of 19 Christmas specials-18 of which were created between 19-that dazzled and occasionally horrified small children. Yet the company is most famous for its holiday specials-often based on popular songs and existing properties-and its innovative use of “Animagic,” which combines stop-motion animation with puppets to create a unique visual style. and Jules Bass, Rankin/Bass created dozens of cartoons from the early 1960s through the 1980s, from ThunderCats to the animated version of J.R.R Tolkien’s The Hobbit. ![]() ![]() From Rudolph to Frosty, no production company has done more to flood the airwaves with Christmas specials than Rankin/Bass Productions. ![]() ![]() ![]() Mirai noticing all the sweets Hibari was eating asks her where she was putting it all. IP 3 binds to IP 3 receptors, channels that release calcium from the endoplasmic reticulum. The other messenger is inositol trisphosphate (IP 3), a molecule that leaves the cell membrane and diffuses within the cytosol. One of these messengers is diacylglycerol (DAG), a molecule that remains within the membrane and activates protein kinase C, which phosphorylates substrate proteins in both the plasma membrane and elsewhere. Phospholipase C splits the PIP 2 into two smaller molecules that each act as second messengers. This lipid component is cleaved by phospholipase C, an enzyme activated by certain G-proteins and by calcium ions. ![]() The two most important messengers of this type are produced from phosphatidylinositol bisphosphate (PIP 2). Remarkably, membrane lipids can also be converted into intracellular second messengers ( Figure 8.7D). Cyclic nucleotide signals are degraded by phosphodiesterases, enzymes that cleave phosphodiester bonds and convert cAMP into AMP or cGMP into GMP.ĭiacylglycerol and IP 3. These cyclic-nucleotide gated channels are ligand-gated (see Chapter 4) they are particularly important in phototransduction and other sensory transduction processes, such as olfaction. In addition, cAMP and cGMP can bind to certain ion channels, thereby influencing neuronal signaling. These enzymes mediate many physiological responses by phosphorylating target proteins, as described in the following section. The most common targets of cyclic nucleotide action are protein kinases, either the cAMP-dependent protein kinase (PKA) or the cGMP-dependent protein kinase (PKG). Once the intracellular concentration of cAMP or cGMP is elevated, these nucleotides can bind to two different classes of targets. Cyclic GMP is similarly produced from GTP by the action of guanylyl cyclase. This enzyme converts ATP into cAMP by removing two phosphate groups from the ATP. Cyclic AMP is produced when G-proteins activate adenylyl cyclase in the plasma membrane. Cyclic AMP is a derivative of the common cellular energy storage molecule, ATP. Another important group of second messengers are the cyclic nucleotides, specifically cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) ( Figure 8.7C). Among the biological signals that activate ryanodine receptors are cytoplasmic Ca 2+ and, at least in muscle cells, depolarization of the plasma membrane.Ĭyclic nucleotides. ![]() A second type of intracellular Ca 2+-releasing channel is the ryanodine receptor, named after a drug that binds to and partially opens these receptors. As the name implies, these channels are regulated by IP 3, a second messenger described in more detail below. One such channel is the inositol trisphosphate ( IP 3) receptor. These intracellular Ca 2+-releasing channels are gated, so they can be opened or closed in response to various intracellular signals. In addition, other channels allow Ca 2+ to be released from the interior of the endoplasmic reticulum into the cytosol. These can be voltage-gated Ca 2+ channels or ligand-gated channels in the plasma membrane, both of which allow Ca 2+ to flow down the Ca 2+ gradient and into the cell from the extracellular medium. The Ca 2+ ions that act as intracellular signals enter cytosol by means of one or more types of Ca 2+-permeable ion channels (see Chapter 4). Such buffers reversibly bind Ca 2+ and thus blunt the magnitude and kinetics of Ca 2+ signals within neurons. Finally, nerve cells contain other Ca 2+-binding proteins-such as calbindin-that serve as Ca 2+ buffers. These organelles can thus serve as storage depots of Ca 2+ ions that are later released to participate in signaling events. In addition to these plasma membrane mechanisms, Ca 2+ is also pumped into the endoplasmic reticulum and mitochondria. Most important in this maintenance are two proteins that translocate Ca 2+ from the cytosol to the extracellular medium: an ATPase called the calcium pump, and an Na + /Ca 2+ exchanger, which is a protein that replaces intracellular Ca 2+ with extracellular sodium ions (see Chapter 4). This steep Ca 2+ gradient is maintained by a number of mechanisms ( Figure 8.7B). ![]() The concentration of Ca 2+ ions outside neurons-in the bloodstream or cerebrospinal fluid, for instance-is several orders of magnitude higher, typically several millimolar (10 –3 M). Ordinarily the concentration of Ca 2+ ions in the cytosol is extremely low, typically 50–100 nanomolar (10 –9 M). ![]() ![]() ![]() It is ideal for those who are new to budgeting. In my opinion, the ideal budget is based on a whole year's worth of income and expense data. That is because the monthly average in these worksheets is calculated based on the number of columns in the row. The above spreadsheets do this for you, but if you haven't completed an entire year, you need to first delete the columns containing the months for which you don't have complete information. To prepare for the final step in making a budget, you need to calculate your average monthly income and average monthly expense for each of the categories. Magazine subscriptions, car repairs, Christmas gifts, and birthday gifts are all examples of expenses that are sprinkled throughout the year, but these must be carefully considered when making a budget. See how the amounts change from month to month? By tracking you expenses from month to month, you will find that you make some purchases only once or twice a year (like going to the dentist). Making a Budget: Recording actual expenses. Below is a snapshot from the budget planner showing how I've recorded my actual expenses for Groceries and Clothing for January through March. The yearly worksheets are great for making a budget because you can record your actual income and expenses for multiple months. At this point, I'd actually start by downloading the yearly personal budget spreadsheet or the family budget planner if you have family obligations. I can't do step 1 for you, but I can help with step 2. When you've gathered all the information, you need to group all your income and expenses into categories. He continues: “For me, storytelling is the fun part of writing music for games and Treyarch spends a lot of time focusing on the narrative in the single-player campaigns. If the music goes away or is not there, and you miss it, then we’re doing our job.” In any case, it’s got to have energy and keep up with the action. Maybe it’s the location or the characters involved that I can latch onto. Or I might have a lot of percussion that feels appropriate for a specific map. “I might do something with high strings or a pad without a lot of conflicting percussion. ![]() But even when the combat is full on, there are ways to heighten the experience for the player, even if the music is playing more of a support role in the background. Certainly, in the immediate adrenaline of an FPS, an immersive cinematic score might not be as much of a priority as compared to the sounds of gunfire, explosions and enemy footsteps. So it must be a surprise then that Wall didn’t continue working on Mass Effect after the second instalment – arguably the series’ peak – but instead leapt into the challenge of composing for Call Of Duty’s Black Ops series, starting with 2012’s Black Ops 2 and continuing with the latest Black Ops Cold War. ![]() ![]() ![]() ![]() Keep an eye out for events and trainings as they become available. recent decades they more often do collaborative work in nonprofit co.
